The synthesizer features dual sections of essential components, including two oscillators, two LFOs, two envelopes, and two filters. Its oscillators draw inspiration from classic analog polysynths, combining multiple voices for a lush, chorused effect through unison mode. Each oscillator includes seven unison voices with adjustable detuning and stereo spread, controlled by a single parameter to create expansive and dynamic textures.

Substance offers extensive modulation options, with LFOs capable of influencing nearly every parameter, such as oscillator frequency, pulse width, filter cutoff, and amplifier volume. LFO-1 provides flexible timing modes including free-running, tempo-synced, and key-synced settings.
